Proximity-Induced Chemical Based Strategy to Reprogram Pathogenic Transcription and Restore Therapeutic Gene Expression

We are developing a TCIP-based transcriptional regulation platform using bivalent small molecules.
In our model system, AP1867-JQ1 physically bridges PPARγ-FKBP^F36V and endogenous BRD4, enhancing PPARγ-driven transcription.
This proximity induction upregulates adipocyte differentiation markers and synergizes with conventional PPARγ ligands.
Based on this principle, we aim to extend chemical proximity-based transcriptional rewiring to rare diseases.
By recruiting transcriptional regulators or chromatin effectors to disease-relevant loci, this strategy may restore beneficial gene expression without permanent genome editing.
